目的:观察脓毒症大鼠在胰岛素联合生长激素治疗前后生长激素受体的变化。方法:将56只造模成功大鼠随机分为七组,即对照组、脓毒症组、胰岛素组、生长激素组、联合组、LY294002组和MG-132组。于给药24 h后留取肝组织标本,检测生长激素受体mRNA和蛋白含量。结果:联合组较生长激素组脓毒症大鼠肝生长激素受体mRNA和蛋白含量增加。阻断PI3K和泛素蛋白酶体途径后,生长激素受体mRNA和生长激素受体蛋白均较联合组表达减少。结论:脓毒症时,胰岛素可通过PI3K信号上调肝细胞的生长激素受体水平,其作用可能与泛素蛋白酶体有关。
Objective: To observe the changes of growth hormone receptor in sepsis rats before and after the combination of insulin and growth hormone. Methods: Fifty-six rats were randomly divided into seven groups: control group, sepsis group, insulin group, growth hormone group, combination group, LY294002 group and MG-132 group. Twenty-four hours after administration, liver samples were collected and the mRNA and protein levels of growth hormone were detected. Results: Compared with the growth hormone group, the hepatic growth hormone receptor mRNA and protein levels increased in sepsis rats. Blocking PI3K and ubiquitin proteasome pathway, growth hormone receptor mRNA and growth hormone receptor protein expression decreased compared with the combined group. Conclusion: In sepsis, insulin can upregulate the growth hormone receptor of hepatocytes via PI3K signaling, which may be related to ubiquitin proteasome.
