目的探讨乳脂球MFG-E8、TGF-β1、TNF-α、ICAM-1等炎症因子与增龄性及颈动脉粥样硬化(CAS)的相关性。方法选取2013年1月~2015年2月期间在新疆医科大学附属中医医院接受体检的健康人群150例,CAS患者150例。采用双抗体夹心ELISA法检测健康人群及CAS患者血清MFG-E8、TGF-β1、TNF-α、ICAM-1等指标。结果健康人群中,随着年龄的增加,MFG-E8、TNF-α、ICAM-1等指标水平呈上升趋势(P<0.05);TGF-β1水平呈下降趋势(P<0.05)。经相关性分析,健康人群MFG-E8、TNF-α、ICAM-1水平与年龄呈正相关,TGF-β1水平与年龄呈负相关。随着CAS病变程度的加重,MFG-E8、ICAM-1水平呈上升趋势(P<0.05);TGF-β1水平呈下降趋势(P<0.05);斑块组和增厚组TNF-α水平高于颈动脉正常组(P<0.05)。斑块组患者MFG-E8、TNF-α、ICAM-1水平与Crouse积分呈正相关,TGF-β1与Crouse积分呈负相关。CAS患者MFGE8、TNF-α、ICAM-1水平之间呈正相关,MFG-E8、TNF-α、ICAM-1与TGF-β1之间呈负相关。结论 MFG-E8、TGF-β1、TNF-α、ICAM-1等指标的增龄性改变与衰老有关,并且与CAS的发病、疾病进展密切相关。
Objective To investigate the relationship between the inflammatory factors such as MFG-E8, TGF-β1, TNF-α, ICAM-1 and age-related carotid atherosclerosis (CAS) in milk fat globules. Methods From January 2013 to February 2015, 150 healthy subjects and 150 CAS patients undergoing physical examination at the Affiliated Chinese Medicine Hospital of Xinjiang Medical University were selected. Double antibody sandwich ELISA was used to detect the serum levels of MFG-E8, TGF-β1, TNF-α and ICAM-1 in healthy people and CAS patients. Results In healthy population, the levels of MFG-E8, TNF-α and ICAM-1 increased with the increase of age (P <0.05), while the levels of TGF-β1 showed a decreasing trend (P <0.05). Correlation analysis showed that the levels of MFG-E8, TNF-α and ICAM-1 in healthy population were positively correlated with age, while the levels of TGF-β1 were negatively correlated with age. The levels of MFG-E8 and ICAM-1 increased with the severity of CAS (P <0.05), while the levels of TGF-β1 decreased (P <0.05). The levels of TNF-α in plaque group and thickening group were higher In the normal carotid artery group (P <0.05). The level of MFG-E8, TNF-α, ICAM-1 in plaque group was positively correlated with Crouse score, while TGF-β1 was negatively correlated with Crouse score. There was a positive correlation between the levels of MFGE8, TNF-α and ICAM-1 in patients with CAS. There was a negative correlation between MFG-E8, TNF-α, ICAM-1 and TGF-β1. Conclusion Age-related changes of MFG-E8, TGF-β1, TNF-α and ICAM-1 are related to aging and are closely related to the pathogenesis and progression of CAS.
