目的:观察hIGF-1工程细胞移植对糖尿病模型小鼠的降血糖作用。方法:用高病毒滴度的病毒上清感染骨髓基质细胞,并植入糖尿病模型小鼠腹腔内,观察糖尿病模型小鼠血糖的变化。结果:稳定表达hIGF-1的骨髓基质细胞腹腔移植后,可见糖尿病小鼠的血糖明显下降(P<0.05)。结论:稳定表达hIGF- 1的骨髓基质细胞对糖尿病动物模型有降糖作用,说明IGF-1基因可作为糖尿病基因治疗的候选基因。
Objective: To observe the hypoglycemic effect of hIGF-1 engineering cell transplantation on diabetic mice. Methods: BMSCs were infected with virus supernatant with high virus titer and implanted into the peritoneal cavity of diabetic mice to observe the changes of blood glucose in diabetic mice. Results: After intraperitoneal transplantation of bone marrow stromal cells stably expressing hIGF-1, the blood glucose of diabetic mice was significantly decreased (P <0.05). CONCLUSION: Bone marrow stromal cells stably expressing hIGF-1 have a hypoglycemic effect on diabetic animal models, indicating that IGF-1 gene may serve as a candidate gene for the gene therapy of diabetes.
