目的探讨丹参对肾间质纤维化(RIF)大鼠肾结缔组织生长因子(CTGF)及细胞周期素依赖性激酶抑制剂(p27)的影响。方法将48只清洁级Wistar雄性大鼠随机分为四组:正常组,假手术组,模型组,丹参治疗组,建立大鼠RIF模型后,给予丹参治疗。取出四组大鼠的左肾组织,用免疫组化法检测CTGF和p27在肾组织的表达。结果丹参能减少CTGF和p27的表达,抑制肾脏细胞异常的肥大,减少细胞外基质(ECM)合成;丹参治疗组与假手术组和正常组大鼠CTGF和p27肾小管阳性细胞数比较,差异有统计学意义(P<0.05)。结论 CTGF和p27在梗阻性RIF发生、发展中起着重要的作用。
Objective To investigate the effect of Salvia miltiorrhiza on renal connective tissue growth factor (CTGF) and cyclin dependent kinase inhibitor (p27) in rats with renal interstitial fibrosis (RIF). Methods Forty-eight clean Wistar male rats were randomly divided into four groups: normal group, sham operation group, model group and Salvia miltiorrhiza treatment group. After establishing rat RIF model, Danshen treatment was given. The left kidney tissues of the four groups of rats were taken out and the expression of CTGF and p27 in the kidney tissues were detected by immunohistochemistry. Results Salvia miltiorrhiza can decrease the expression of CTGF and p27, inhibit the abnormal hypertrophy of renal cells and decrease the synthesis of extracellular matrix (ECM). Compared with sham operation group and normal group, the number of tubule positive cells in CTGF and p27 Statistical significance (P <0.05). Conclusion CTGF and p27 play an important role in the occurrence and development of obstructive RIF.
