目的:探讨表皮生长因子(EGF)、转化生长因子α和β1(TGFα,TGFβ1) 对肺泡Ⅱ型细胞(ATⅡ) 磷脂分泌功能及细胞形态( 肥大) 的影响。方法:原代培养的成年大鼠ATⅡ中,加入生长因子作用48 h,应用化学测定法测定培养介质中总磷脂含量,采用图像分析技术测量细胞体积参数。结果:EGF、TGFα均可使总磷脂含量增加,而TGFβ1 则使总磷脂含量降低;随EGF、TGFα浓度增加,ATⅡ平均直径(DQ) 、体积(VQ) 和表面积(SQ) 均逐渐增大,TGFβ1 对细胞体积变化无明显影响;TGFα和TGFβ1 同时加入培养体系中,总磷脂含量及细胞体积均无明显改变。结论:EGF、TGFα能促进ATⅡ磷脂分泌和细胞肥大;TGFβ1 对TGFα的作用有抑制效应。
Objective: To investigate the effects of epidermal growth factor (EGF), transforming growth factor α (TGFβ1) and transforming growth factor β1 (TGFβ1) on the phospholipid secretion and cell morphology (hypertrophy) of alveolar type Ⅱ cells. Methods: Primary cultured rat AT Ⅱ was treated with growth factor for 48 h. The content of total phospholipids in the medium was determined by chemical assay. The cell volume parameters were measured by image analysis. Results: The total phospholipid content was increased by EGF and TGFα, while the total phospholipid content by TGFβ1 was decreased. With the increase of EGF and TGFα concentration, the average diameter (DQ), volume (VQ) and surface area (SQ) TGFβ1 had no significant effect on cell volume change; TGFα and TGFβ1 were added into the culture system at the same time, and the total phospholipid content and cell volume did not change significantly. Conclusion: EGF and TGFα can promote the secretion of AT Ⅱ phospholipid and cell hypertrophy. The effect of TGFβ1 on TGFα is inhibited.
