目的 探讨多药耐药逆转剂是否增加化疗药物对正常造血干细胞的毒性。方法 采用甲基纤维素半固体培养方法,培养正常人粒细胞- 巨噬细胞克隆形成单位(CFU-GM)。测定去甲柔红霉素对CFU-GM 的抑制率。将多药耐药逆转剂MS-209及去甲柔红霉素与骨髓细胞共同短期作用后,了解去甲柔红霉素对CFU-GM 抑制率的变化。结果 去甲柔红霉素对CFU-GM 的平均抑制率为29.6% ,在MS-209作用下,其抑制率增加为43.4% ,有显著性差异(P< 0.05)。结论 多药耐药逆转剂MS-209 增加了化疗药物对造血干细胞的毒性。提示该类药物在临床应用中可能对造血系统具有一定副作用。
Objective To investigate whether multidrug resistance reversal agents increase the toxicity of chemotherapeutic drugs to normal hematopoietic stem cells. Methods Normal human granulocyte-macrophage colony forming units (CFU-GM) were cultured using methylcellulose semi-solid culture method. The inhibition rate of norfomycin to CFU-GM was measured. After a short-term effect of the multidrug resistance reversal agent MS-209 and norfomycin and bone marrow cells, the change of the inhibitory rate of CFU-GM by nordo daunorubicin was understood. Results The average inhibition rate of norfloxacin on CFU-GM was 29.6%. Under the effect of MS-209, the inhibition rate was increased by 43.4% with significant difference (P<0.05). Conclusion MS-209, a multidrug resistance reversal agent, increases the toxicity of chemotherapeutic drugs to hematopoietic stem cells. It is suggested that this type of drug may have some side effects on the hematopoietic system in clinical applications.
