Estrogen receptor alpha (ERa), a member of the nuclear receptor superfamily, exerts its biological functions predominantly in a ligand-dependent manner. ERα plays a role in female reproductive system development and affects the proliferation and differentiation of endometrium. Studies confirm that ERα-mediated gene transcription plays an important role in carcinogenesis of type I endometrium. In addition, estrogen receptor β (ERβ) or estrogen-related receptor (ERR) is also involved in this process and has a functional relationship with ERα. In this review, we mainly reviewed ERα-mediated transcriptional regulation of transcriptional regulators, important downstream target genes of ERα and their roles in endometrial cancer, and provided new ideas for the early detection and treatment of endometrial cancer.
