目的:探讨牛蒡根水提物对血管内皮细胞损伤的保护作用及其机制。方法:用脂多糖(LPS)诱导血管内皮细胞损伤模型,随机分组为空白对照组,模型组和牛蒡根水提物低、中、高浓度组。用酶联免疫吸附试验(ELISA)测定细胞培养上清中人细胞间黏附分子-1(ICAM-1)、人肿瘤坏死因子-α(TNF-α)、人基质金属蛋白酶-1(MMP-1)、人超敏C反应蛋白(hs-CRP)水平;用PCR检测细胞Toll样受体4(TLR4)、核因子(NF-κB)的抑制因子(I-κB)的表达水平。结果:与模型组比较,药物治疗组各组炎性因子ICAM-1、TNF-α、MMP-1及hsCRP水平均降低(P<0.05),TLR4 m RNA表达水平降低(P<0.05),I-κB m RNA表达水平增高(P<0.05)。结论:牛蒡根水提物可能是通过抑制细胞TLR4、NF-κB的表达,从而抑制炎性因子ICAM-1、TNF-α、MMP-1及hs-CRP等的产生继而保护细胞的。
Objective: To investigate the protective effect of water extract of Arctium lappa L. on vascular endothelial cell injury and its mechanism. Methods: The model of vascular endothelial cell injury induced by lipopolysaccharide (LPS) was randomly divided into blank control group, model group and low, middle and high concentration of Arctium lappa extract. The expression of ICAM-1, TNF-α, MMP-1 (MMP-1) in cell culture supernatants were determined by enzyme-linked immunosorbent assay (ELISA) ) And hs-CRP were detected by flow cytometry. The expression of TLR4 and NF-κB inhibitor I-κB was detected by PCR. Results: Compared with the model group, the levels of ICAM-1, TNF-α, MMP-1 and hsCRP were decreased (P <0.05), the expression of TLR4 mRNA was decreased (P <0.05) -κB m RNA expression increased (P <0.05). Conclusion: The water extract of burdock root may protect the cells by inhibiting the expression of TLR4 and NF-κB, thereby inhibiting the production of inflammatory factors such as ICAM-1, TNF-α, MMP-1 and hs-CRP.
