目的:建立一种快速筛选和评价抗菌活性物质的方法,并将其应用于海洋抗菌活性物质的筛选。方法:在对生物样品的提取过程中引入加速溶剂萃取技术(ASE),同时采用刃天青作为指示剂间接判断不同提取物样品的抗菌特性。结果:建立了一种快速筛选抗菌药物的方法,并发现白棘三列海胆、梅氏长海胆、面包海星、秀丽节蛇尾的乙酸乙酯提取物对实验菌生长均具有较强抑制作用。结论:该方法克服了常规的纸片琼脂扩散法、试管肉汤稀释法等作为抗菌药物筛选方法的不足,可使数量庞大的生物活性物质中抗菌药物的高通量筛选与评价成为可能。
Objective: To establish a rapid screening and evaluation of antimicrobial active substances, and apply it to the screening of marine antimicrobial active substances. Methods: Accelerated solvent extraction (ASE) was introduced in the extraction of biological samples, and the antibacterial properties of different extracts were determined indirectly by using resazurin as an indicator. Results: A rapid screening method for antimicrobial agents was established. It was found that the ethyl acetate extract of three kinds of white sea urchin, sea buckthorn, seaweed bread, star anise snail tail had a strong inhibitory effect on the growth of experimental bacteria. Conclusion: This method overcomes the shortcomings of conventional paper agar diffusion method and tube broth dilution method as screening methods of antibacterial drugs, which can make high-throughput screening and evaluation of antibacterial drugs in a large number of bioactive substances possible.
