益肾解毒汤对单侧输尿管梗阻大鼠模型肾组织氧化应激反应的干预作用

目的研究益肾解毒汤对单侧输尿管梗阻(UUO)大鼠肾组织氧化应激的干预作用。方法用UUO法建立慢性肾衰竭大鼠模型,SD雄性大鼠随机分为假手术组、模型组、益肾解毒汤组和科素亚组,于造模后7d、14d各组随机选取10只检测血肌酐(Scr),尿素氮(BUN),24h尿蛋白定量(24h Upro),肾组织匀浆超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)。结果与假手术组比较,模型组肾组织SOD、GSH明显降低(P<0.01),Scr、BUN、24h Upro、MDA明显升高(P<0.01);益肾解毒汤组与模型组比较,肾组织SOD、GSH明显升高(P<0.01),Scr、BUN、24h Upro、MDA明显下降(P<0.05);益肾解毒汤组与科素亚组比较,肾组织SOD、GSH明显升高(P<0.05),Scr、BUN、24h Upro、MDA明显下降(P<0.05)。结论在肾间质纤维化的整个发展过程中,都有氧化应激反应的参与,益肾解毒汤能有效减轻慢性肾衰竭大鼠的氧化应激反应,进而可能起到延缓肾间质纤维化的作用。 Objective To study the intervention effect of Yishen Jiedu Decoction on oxidative stress in renal tissue of unilateral ureteral obstruction (UUO) rats. Methods Rat models of chronic renal failure were established by UUO method. SD male rats were randomly divided into sham-operated group, model group, Yishen Jiedu decoction group and co-sub-group. Ten rabbits were randomly selected from each group on the 7th day and the 14th day after modeling. Serum creatinine (Scr), blood urea nitrogen (BUN), 24 h urine protein (24 h), renal tissue homogenate superoxide dismutase (SOD), malondialdehyde (MDA) and glutathione (GSH) were measured. Results Compared with the sham operation group, the levels of SOD and GSH in the model group were significantly decreased (P <0.01), while the levels of Scr, BUN, 24h and Upro and MDA in the model group were significantly increased (P <0.01) The levels of SOD and GSH were significantly increased (P <0.01), Scr, BUN, 24h, Upro and MDA were significantly decreased in the Yishen Jiedu Decoction group (P <0.05) P <0.05). Scr, BUN, 24h Upro, MDA decreased significantly (P <0.05). Conclusions In the whole process of renal interstitial fibrosis, there is the participation of oxidative stress reaction. Yishen Jiedu Decoction can effectively reduce the oxidative stress response in rats with chronic renal failure, and thus may delay renal interstitial fibrosis Role.
