目的 研究血管内皮生长因子(VEGF)、诱导型一氧化氮合酶(iNOS)和内皮型一氧化氮合酶(eNOS)在颜面皮肤癌中表达的相关性;探讨三者与颜面皮肤癌血管生成和临床病理特征的关系;研究一氧化氮(NO)和 VEGF的相互作用及 NO在 VEGF促肿瘤生长中的作用机制。方法 应用免疫组织化学方法检测47例颜面皮肤癌手术切除标本 VEGF、iNOS和 eNOS的表达,Ⅷ因子相关抗原(FⅧRAg)血管内皮细胞特异性染色计数肿瘤微血管密度(MVD)。结果 (1)32例颜面皮肤癌组织表达VEGF,30例表达iNOS,35例表达eNOS;(2)VEGF与iNOS的表达呈正相关,与eNOS的表达无相关;(3)VEGF、iNOS的表达与颜面皮肤癌MVD呈正相关, eNOS的表达与颜面皮肤癌MVD的差异无显著性意义;(4)VEGF的表达与颜面皮肤癌淋巴结转移和肿瘤分化程度及肿瘤浸润深度呈正相关;与临床分期无关。iNOS表达与颜面皮肤癌的分化程度及临床分期和有无淋巴结转移及肿瘤浸润深度正相关;eNOS表达与颜面皮肤癌临床分期和分化程度及有无淋巴结转移及肿瘤浸润深度均无关。结论 (1)iNOS对 VEGF的生成和发挥作用的过程有重要影响;(2)MVD随着VEGF和iNOS表达的增强而增加,说明两者对颜面皮肤癌血管生成具有促进作用。
Objective To investigate the correlation between the expression of vascular endothelial growth factor (VEGF), inducible nitric oxide synthase (iNOS) and endothelial nitric oxide synthase (eNOS) in facial skin cancer, and to explore the relationship between them and facial skin angiogenesis And the clinicopathological features of the tumor; to study the interaction between nitric oxide (NO) and VEGF and the mechanism of NO in tumor growth. Methods Immunohistochemistry was used to detect the expression of VEGF, iNOS and eNOS in 47 surgically resected skin cancer specimens. The tumor microvessel density (MVD) was counted by Vascular endothelial cell specific staining of factor Ⅷ related antigen (FⅧRAg). Results (1) 32 cases of facial skin cancer expressed VEGF, 30 cases expressed iNOS and 35 cases expressed eNOS. (2) The expression of VEGF and iNOS was positively correlated with the expression of eNOS. (3) The expression of VEGF, iNOS and There was a positive correlation between MVD in facial skin cancer and MVD in facial skin cancer. (4) The expression of VEGF was positively correlated with lymph node metastasis, tumor differentiation and depth of tumor invasion in facial skin cancer, but not with clinical stage. The expression of iNOS was positively correlated with the degree of differentiation of facial skin cancer, clinical stage and lymph node metastasis and the depth of tumor invasion. There was no correlation between the expression of iNOS and the clinical stages and differentiation of facial skin cancer with or without lymph node metastasis and the depth of tumor invasion. Conclusions (1) iNOS plays an important role in the process of VEGF production and expression. (2) MVD increases with the expression of VEGF and iNOS, indicating that both of them can promote angiogenesis of facial skin cancer.
