[目的]观察硫酸镁预处理及硫酸镁治疗对兔全脑缺血后的神经保护作用。[方法]40只兔随机分为4组:对照组、缺血组、镁预处理组和镁治疗组,每组10只。阻断4血管(双侧颈总动脉及椎动脉),诱导全脑缺血,缺血时间为6min。测定缺血再灌注30min海马氨基酸含量。检测缺血再灌注3d海马CA1区神经元密度及凋亡神经元密度。[结果](1)镁预处理组及镁治疗组海马CA1区神经元密度显著高于缺血组(P﹤0.01),凋亡神经元密度显著低于缺血组(P﹤0.01)。(2)镁预处理组及镁治疗组海马天冬氨酸和甘氨酸含量显著低于缺血组(P﹤0.01);而γ-氨基丁酸含量显著高于缺血组(P﹤0.05)。(3)镁预处理组与镁治疗组比较各指标差异无统计学意义(P﹥0.05)。[结论]硫酸镁预处理及硫酸镁治疗对兔全脑缺血有显著的神经保护作用。
[Objective] To observe the neuroprotective effects of magnesium sulfate preconditioning and magnesium sulfate on global cerebral ischemia in rabbits. [Method] Forty rabbits were randomly divided into 4 groups: control group, ischemia group, magnesium pretreatment group and magnesium treatment group, 10 in each group. Blocked 4 blood vessels (bilateral common carotid artery and vertebral artery), induced global cerebral ischemia, ischemic time was 6min. Determination of hippocampal amino acid content 30 min after ischemia reperfusion. The neuron density and the density of apoptotic neurons in the hippocampal CA1 hippocampus after ischemia / reperfusion were measured. [Results] (1) The density of neurons in hippocampal CA1 region of magnesium pretreatment group and magnesium treatment group was significantly higher than that of ischemia group (P <0.01), and the density of apoptotic neurons was significantly lower than that of ischemia group (P <0.01). (2) The content of aspartate and glycine in the hippocampus of magnesium pretreatment group and magnesium treatment group was significantly lower than that of ischemia group (P <0.01), while the content of γ-aminobutyric acid was significantly higher than that of ischemia group (P <0.05). (3) The magnesium pretreatment group and magnesium treatment group showed no significant difference (P> 0.05). [Conclusion] Magnesium sulfate preconditioning and magnesium sulfate have significant neuroprotective effects on global cerebral ischemia in rabbits.
