肝巨噬细胞在肝纤维化的损伤和修复阶段发挥着不同的作用,具有功能异质性。早期研究表明肝巨噬细胞的这种功能异质性可能与其具有不同的活化模式密切相关。然而,近年来关于巨噬细胞起源的研究打破了免疫学教科书中传统理论:即成体组织定居巨噬细胞来源于胚胎时期的卵黄囊。因而肝巨噬细胞的功能异质性是否和其起源相关,是目前巨噬细胞领域研究的热点。本文拟从巨噬细胞功能、活化和起源异质性,结合不同肝纤维化模型中巨噬细胞作用的研究进展和靶向巨噬细胞治疗的应用三个方面进行总结,有望为临床开展肝纤维化的细胞治疗提供新思路和新策略。
Liver macrophages play different roles in the stage of injury and repair of liver fibrosis with functional heterogeneity. Early studies have shown that this functional heterogeneity of liver macrophages may be closely related to its different activation patterns. However, recent studies on the origin of macrophages have broken the conventional wisdom in textbooks of immunology that the adult tissue colonizing macrophages is derived from the yolk sac of the embryonic period. Therefore, whether the functional heterogeneity of hepatic macrophages is related to its origin is the hotspot in the field of macrophages. This article intends to summarize three aspects of macrophage function, activation and origin of heterogeneity, combined with the research progress of macrophages in different liver fibrosis models and the application of targeted macrophages treatment, which is expected to be useful for clinical development of liver fibrosis Cell therapy provides new ideas and new strategies.
