目的 为临床探索一种更简便安全的下肢动脉缺血性疾病的基因治疗途径。方法 建立兔后肢缺血动物模型 ,将体外构建的重组质粒 pSV VEGF16 5注射于缺血肌群 ,30d后行动脉造影并切取标本 ,测定毛细血管密度和毛细血管 /肌肉数比值。结果 经 pSV VEGF16 5治疗后 ,缺血肢体毛细血管密度和毛细血管 /肌肉数比值显著增加 ,小腿部位比大腿部位增加明显 ;动脉造影显示部分缺血肢体侧支动脉明显增多。结论 肌肉内原位注射 pSV VEGF16 5是一种简单有效的治疗基因转染途径 ,能显著增加缺血肢体侧支血管形成。
Objective To explore a more simple and safe method of gene therapy for lower extremity arterial ischemic disease. Methods The animal model of hindlimb ischemia in rabbits was established. The recombinant plasmid pSV VEGF165 was injected into the ischemic muscle group. The arterial angiography was performed 30 days later. The capillary density and capillary / muscle ratio were measured. Results After treatment with pSV VEGF165, the capillary density and capillary / muscle ratio of ischemic limbs increased significantly, and the position of the lower leg increased significantly compared with that of the thigh. Angiography showed that the number of collateral arteries in some ischemic limbs increased significantly. Conclusion Intramuscular injection of pSV VEGF16 5 is a simple and effective approach to gene transfection, which can significantly increase the collateral vessels in ischemic limbs.
