目的:观察强筋壮骨祛风合剂对腰椎间盘退变水通道蛋白1(AQP1)和水通道蛋白5(AQP5)表达的干预作用。方法:将新西兰大白兔腰椎穿刺建立腰椎间盘退变模型,用RT-PCR及免疫组化检测AQP1和AQP5在椎间盘组织中的表达,观察模型大白兔给药6周后的变化。结果:1)RT-PCR检测:(1)各组AQP1表达比较,假手术组与空白对照组差异无统计学意义(P>0.05),模型空白组、腰痛宁组、强筋壮骨祛风合剂组与空白对照组差异有统计学意义(P<0.05),空白对照组、假手术组、腰痛宁组、强筋壮骨祛风合剂组与模型空白组差异有统计学意义(P<0.05)。(2)各组AQP5表达比较,假手术组与空白对照组差异无统计学意义(P>0.05),模型空白组、腰痛宁组、强筋壮骨祛风合剂组与空白对照组差异有统计学意义(P<0.05),空白对照组、假手术组、腰痛宁组、强筋壮骨祛风合剂组与模型空白组差异有统计学意义(P<0.05)。2)免疫组化试剂检测:(1)各组AQP1表达比较,造模成功后,模型空白组椎间盘上AQP1阳性面积与空白对照组、假手术组差异有统计学意义(P<0.05),空白对照组和假手术组差异无统计学意义(P>0.05),其余组之间差异均有统计学意义(P<0.05)。(2)各组AQP5表达比较,造模成功后,模型空白组椎间盘上AQP5阳性面积与空白对照组、假手术组差异有统计学意义(P<0.05),空白对照组和假手术组差异无统计学意义(P>0.05),其余组之间差异均有统计学意义(P<0.05)。结论:强筋壮骨祛风合剂对AQP1及AQP5的调节作用是其疗效机制之一,具有改善腰椎间盘退变的作用。
Objective: To observe the intervention effect of “Qiangjin Zhuanggu Qufeng Mixture” on expression of aquaporin 1 (AQP1) and aquaporin 5 (AQP5) in lumbar intervertebral disc. Methods: The lumbar intervertebral disc degeneration model was established in New Zealand white rabbits by lumbar puncture. The expression of AQP1 and AQP5 in the intervertebral disc was detected by RT-PCR and immunohistochemistry. The changes of the model rabbits after 6 weeks were observed. Results: 1) RT-PCR: (1) The expression of AQP1 in each group was not significantly different between the sham operation group and the blank control group (P> 0.05), the model blank group, the Yaotongning group, There was a significant difference between the control group and the blank control group (P <0.05), and there was significant difference between the blank control group, sham operation group, Yaotongning group, ). (2) The expression of AQP5 in each group showed no significant difference between the sham operation group and the blank control group (P> 0.05), there was statistical difference between the model blank group, the Yaotongning group, the strong gluten, the bone-strengthening and Qufeng Mixture group and the blank control group (P <0.05). There was significant difference between the blank control group, the sham operation group, the Yaotongning group, the Qiangjin Zhuanggu Qugan Mixture group and the model blank group (P <0.05). 2) Immunohistochemical detection: (1) Comparison of AQP1 expression between groups showed that the positive area of AQP1 in the model blank group was significantly different from the blank control group and sham operation group (P <0.05) There was no significant difference between the control group and the sham operation group (P> 0.05). The differences between the other groups were statistically significant (P <0.05). (2) Compared with the control group and the sham operation group, the AQP5 positive area in the model blank group was statistically significant (P <0.05), and there was no significant difference between the blank control group and sham operation group Statistical significance (P> 0.05), the difference between the other groups were statistically significant (P <0.05). CONCLUSION: The effect of QGZQQQJ on AQP1 and AQP5 is one of its therapeutic mechanisms, which has the effect of improving lumbar disc degeneration.
