目的分别比较热休克蛋白60(HSP60)在多西紫杉醇(Docetaxel)诱导前列腺癌PC-3细胞凋亡及耐药前后表达的差异,并探讨其意义。方法采用20μmol/L Docetaxel诱导PC-3细胞凋亡,后采用逐渐加量的方式培养PC-3细胞Docetaxel耐药株;采用双向荧光差异凝胶电泳(DIGE)技术定量筛选未处理组PC-3细胞与凋亡PC-3细胞,及PC-3细胞敏感株与耐药株的差异蛋白,并采用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)技术对其进行成分鉴定,进而检测HSP60的表达。结果 HSP60在Docetaxel诱导PC-3凋亡细胞中表达上调,在Docetaxel耐药株中表达下调。结论 HSP60在PC-3凋亡细胞中高表达,在Docetaxel耐药株中的低表达,提示其可能与PC-3细胞的凋亡及耐药有重要关系。
Objective To compare the difference of HSP60 expression in PC-3 cells induced by Docetaxel before and after drug-resistant, and to explore its significance. Methods PC-3 cells were induced by Docetaxel (20μmol / L). Docetaxel-resistant strains of PC-3 cells were cultured with increasing doses. The levels of PC-3 in PC-3 cells were quantified by two-dimensional fluorescence difference gel electrophoresis The differentially expressed proteins of PC-3 cells, PC-3 cells and PC-3 cells were detected by MALDI-TOF-MS. HSP60 expression. Results HSP60 was up-regulated in Docetaxel-induced PC-3 apoptotic cells and down-regulated in Docetaxel-resistant cells. Conclusions HSP60 is highly expressed in PC-3 apoptotic cells and low in Docetaxel-resistant strains, suggesting that HSP60 may play an important role in the apoptosis and drug resistance of PC-3 cells.
