目的 :探讨低浓度CO对大鼠主动脉平滑肌细胞迁移的影响。方法 :体外培养大鼠主动脉平滑肌细胞 ,用低浓度CO处理 2 4、48、72h ,检测平滑肌细胞迁移情况。结果 :2 4、48、72h后未加处理的对照组大鼠主动脉平滑肌细胞迁移距离逐渐增加 ,与 2 4h组比差异显著 (P <0 0 0 1)。 2 0× 10 -6、5 0× 10 -6、80× 10 -6CO处理细胞 2 4h后 ,细胞迁移距离逐渐下降 ,2 0× 10 -6CO组与对照组差异不显著 (P <0 0 5 ) ,其余两组均显著低于对照组 (P <0 0 1)。 2 0× 10 -6、5 0× 10 -6、80× 10 -6CO处理细胞 48、72h后 ,其迁移距离也显著低于对照组 (P <0 0 1)。结论 :大鼠主动脉平滑肌细胞自身存在迁移情况 ,培养时间愈长 ,迁移距离愈大。低浓度CO可以显著抑制平滑肌细胞的迁移 ,随着浓度增加 ,抑制作用增强。
Objective: To investigate the effect of low concentration CO on the migration of aortic smooth muscle cells in rats. Methods: Aortic smooth muscle cells were cultured in vitro and treated with low concentration of CO for 2, 48, 72 hours to detect the migration of smooth muscle cells. RESULTS: After 2, 48, and 72 hours, the migration of aortic smooth muscle cells increased gradually in untreated control group, which was significantly different from that in 24 hours (P <0.01). After 2 × 10 -6, 5 × 10 -6 and 80 × 10 -6 CO 2 h, the cell migration distance decreased gradually, but the difference was not significant between the 20 × 10 -6 CO group and the control group (P <0.05 ), The other two groups were significantly lower than the control group (P <0.01). The migration distance of cells treated with 2 × 10 -6, 5 × 10 -6 and 80 × 10 -6 CO for 48 and 72 h was also significantly lower than that of the control group (P <0.01). Conclusion: The aorta smooth muscle cells of rats have their own migration, the longer the culture time, the greater the migration distance. Low concentrations of CO can significantly inhibit the migration of smooth muscle cells, with increasing concentration, inhibition increased.
