目的探讨lncRNA TUG1(taurine up-regulated 1)与冠心病的关系,研究TUG1对脐静脉内皮细胞(HUVECs)功能的影响。方法采用real-time PCR检测冠心病病例对照外周血单个核细胞(PBMCs)中TUG1的表达。利用siRNA敲低HUVECs内TUG1表达水平,并检测对细胞凋亡及IL-8表达水平的影响。建立并应用低氧和TNF-α刺激的内皮细胞损伤模型,探讨TUG1在内皮细胞功能紊乱中的作用。结果冠心病患者PBMCs样本中TUG1表达显著高于对照组(P<0.001)。敲低TUG1显著抑制正常培养及低氧或TNF-α刺激诱导的内皮细胞凋亡(P<0.05)。敲低TUG1并予以低氧或TNF-α刺激,IL-8的表达明显下降(P<0.001)。结论 LncRNA TUG1在冠心病患者中出现异常高表达。体外实验结果显示,敲低TUG1能抑制内皮细胞凋亡,降低IL-8的表达水平。提示TUG1可能影响血管内皮细胞功能,参与冠心病的发生发展过程。
Objective To investigate the relationship between lncRNA TUG1 and coronary heart disease (CHD) and to investigate the effect of TUG1 on the function of human umbilical vein endothelial cells (HUVECs). Methods Real-time PCR was used to detect TUG1 expression in peripheral blood mononuclear cells (PBMCs) of patients with coronary heart disease. The expression of TUG1 in HUVECs was knocked down by siRNA and the effect on the apoptosis and IL-8 expression was also examined. Establishment and application of hypoxic and TNF-α-stimulated endothelial cell injury model to explore the role of TUG1 in endothelial dysfunction. Results The expression of TUG1 in PBMCs of patients with coronary heart disease was significantly higher than that of the control group (P <0.001). Knockdown of TUG1 significantly inhibited endothelial cell apoptosis induced by hypoxia or TNF-α stimulation (P <0.05). TUG1 knockdown and hypoxia or TNF-α stimulation, IL-8 expression was significantly decreased (P <0.001). Conclusion LncRNA TUG1 is abnormally expressed in patients with coronary heart disease. In vitro experiments show that knockdown of TUG1 can inhibit endothelial cell apoptosis and reduce the expression of IL-8. Tip TUG1 may affect the function of vascular endothelial cells, involved in the occurrence and development of coronary heart disease.
