本研究通过对比不同凝胶组成成分及电泳条件,建立了非同位素PCR-SSCP方法以检测p53基因第4外显子第72密码子的多态性。对35例正常组织DNA的研究结果显示,p53基因这位点的杂合率和纯合率分别为48.6%(CCC/CGC),45.7%(CGC/CGC)和5.7%(CCC/CCC).本法简单、快速,能有效地检测基因多态性,并可进一步应用于对肿瘤细胞p53基因杂合性缺失的分析。
In this study, a non-isotopic PCR-SSCP method was established to compare the polymorphism of the 72nd codon of exon 4 of p53 gene by comparing different gel components and electrophoresis conditions. The results of the 35 normal tissue DNA studies showed that the heterozygous and homozygous rates of the p53 gene were 48.6% (CCC/CGC), 45.7% (CGC/CGC), and 5.7% respectively. (CCC/CCC). This method is simple, rapid, and can effectively detect genetic polymorphisms, and can be further applied to the analysis of the loss of heterozygosity of p53 gene in tumor cells.
