目的 :研究EGR 1、c fos、cyclinD13种基因mRNA及其相应蛋白在人食管癌癌变过程中的表达水平及其相关性 ,并探讨其与食管癌的发生、发展及生物学行为的关系。方法 :应用原位杂交法检测 47例食管鳞状细胞癌及其癌旁、上切缘中 3种基因的mRNA、免疫组化SABC法检测蛋白表达水平。结果 :3种基因原位杂交、免疫组化阳性表达产物均分别定位于胞浆及胞核中。EGR 1mRNA、c fosmRNA、cyclinD1mRNA及蛋白分别在食管粘膜上皮非典型增生和鳞癌中呈高表达。表现为非典型增生粘膜中EGR 1、c fos表达高 ,cyclinD1表达低 ,进展为癌后 ,EGR 1表达下降 ,c fos表达持续 ,cyclinD1表达则变高 ;3种基因mRNA与其相应蛋白在各自高表达病变类型中表达具有一致性。结论 :食管癌癌变过程各病变类型EGR 1,c fos,cyclinD1表达不同 ,癌基因和抑癌基因的相互作用、相互制约将决定食管癌癌变过程的发生、发展及转归。
Objective: To study the expression levels and correlation of 13 mRNAs of EGR 1, c fos, and cyclin D in human esophageal cancers and their relationship with the occurrence, development and biological behavior of esophageal cancer. Methods: In situ hybridization was used to detect the mRNA of 47 genes in 47 cases of esophageal squamous cell carcinoma and its paraneoplastic and superior margins. Immunohistochemical SABC method was used to detect the protein expression. Results : Three kinds of gene in situ hybridization and immunohistochemical positive expression products were located in the cytoplasm and nucleus, respectively. EGR 1 mRNA, c fos mRNA, cyclinD 1 mRNA and protein were highly expressed in esophageal mucosa epithelial dysplasia and squamous cell carcinoma, respectively. The expression of EGR 1 and c fos in atypical hyperplasia mucosa was high, cyclinD1 expression was low, and after progression to cancer, the expression of EGR 1 decreased, the expression of c fos persisted, and the expression of cyclinD 1 became higher; the mRNAs of the three genes and their corresponding proteins were high There is consistency in expression in the expression lesion type. Conclusion : The expression of EGR 1, c fos and cyclinD1 in cancerous process of esophageal cancer is different. The interaction and mutual restraint between oncogene and tumor suppressor gene will determine the occurrence, development and prognosis of carcinogenesis of esophageal cancer.
