目的:研究槲皮素(Quercetin,QUE)对培养人视网膜色素上皮(retinalpigmentepithelium,RPE)细胞增殖周期的影响。方法:用流式细胞仪测定细胞周期,观察不同浓度(200、100、50、1μmol/L)的QUE作用于RPE细胞48小时;以及QUE200μmol/L在不同作用时间(24~120小时),分别单独或与EGF共同对RPE细胞周期的影响;QUE连续作用于BPE细胞120小时后洗脱QUE,继续培养RPE细胞120小时内的细胞周期变化;QUE对RPE的细胞毒作用。结果:随QUE浓度增大,RPE细胞的S%减小;200μmol/LQUE在120小时内持续抑制RPE细胞进入S期;QUE对EGF促进RPE细胞进入S期的抑制效应更强;所有QUE作用组的细胞活力均在85%以上,移去QUE后,RPE细胞能重新进入细胞周期继续增殖。结论:QUE抑制RPE细胞从GO/GI期进入S期,具剂量依赖效应,在120小时内QUE能持续有效抑制RPE细胞进入S期,抑制效应具一定的可逆性,QUE对RPE细胞无细胞毒作用。

CONCLUSION: QUE inhibits the RPE cells from entering the S phase during the GO / GI phase in a dose-dependent manner. QUE can effectively and effectively inhibit RPE cells entering the S phase within 120 hours, with a certain reversibility of the inhibitory effect. QUE has no cytotoxic effect on RPE cells effect.
