利用克隆系NG108-15细胞在膜片箝全细胞(whole-cellclamprecording)记录下,观察了国产TTX单克隆抗体8A5拮抗TTX对跨膜钠电流的作用。结果表明,8A5有效地抑制TTX的作用:(1)在标准灌流液中加入1μmol/LTTX,Na+流立即被完全阻断,复加1μmol/L8A5于灌流液,可使Na+流迅速恢复至对照水平;(2)若先以含1μmol/L8A5的溶液灌流细胞,再更置为含1μmol/LTTX的标准液,Na+流只有部分被抑制。
Using the clonal NG108-15 cells in whole-cell clamp recording, the effect of TTX monoclonal antibody 8A5 antagonizing TTX on transmembrane sodium current was observed. The results showed that 8A5 effectively inhibited the action of TTX: (1) 1μmol / LTTX was added into the standard perfusate solution, and the Na + flux was blocked immediately. The addition of 1μmol / L 8A5 to the perfusate rapidly restored the Na + flux to the control level ; (2) If the cells were perfused with a solution containing 1μmol / L 8A5 and then replaced with a standard solution containing 1μmol / L of LTT, only part of the Na + flow was inhibited.
