目的建立同时测定寿胎丸煎煮液中金丝桃苷、槲皮素和山奈酚3种黄酮类化合物含量的分析方法,探讨流速、流动相、检测波长对3种化合物分离检测的影响。方法采用固相萃取技术,结合超高效液相色谱分析手段,考察3种黄酮类化合物的色谱分离行为,优化固相萃取条件,在寿胎丸煎煮液样品中分别添加0.05、0.2、0.5 mg/L3个水平的混合标准溶液进行加标回收实验。结果平均回收率为90.0%~100.8%,相对标准偏差为1.4%~4.4%。结论超高效液相色谱方法检测限低、灵敏度高、重现性好,可用于寿胎丸煎煮液中黄酮类化合物的检测。
OBJECTIVE To establish a method for simultaneous determination of three flavonoids in hypericin, quercetin and kaempferol in Shoutai pill, and to study the effects of flow rate, mobile phase and detection wavelength on the separation and detection of three compounds. Methods Solid phase extraction (SPE) and ultra performance liquid chromatography (HPLC) were used to investigate the chromatographic separation behavior of three flavonoids. The conditions of solid phase extraction were optimized. The samples of Shoutai Wan Decoction were respectively added with 0.05, 0.2 and 0.5 mg / L3 levels of mixed standard solution for spike recovery experiments. Results The average recovery was 90.0% ~ 100.8% with relative standard deviation of 1.4% ~ 4.4%. Conclusion Ultra-performance liquid chromatography has the advantages of low detection limit, high sensitivity and good reproducibility. It can be used for the determination of flavonoids in Shoutai pill decoction.
