本文采用不同免疫组织化学方法鉴定抗人结肠癌3 E_4单抗(鼠源)的特异性。共做标本212,发现与胰酶消化处理标本荧光染色效果较好,能排除非特异性抗原干扰。根据该法鉴定,3E_4对结肠癌阳性率为100%,对其他腺癌阳性率(胰、肺、胃)为80%,正常结肠阳性率为0%。在结肠癌癌巢分布密度以周膜、界膜最高,分布率为100%,结肠癌旁分市区为隐窝上部,正常结肠组织无交叉反应,显示3E_4单抗为抗人结肠相关性抗体。根据封闭试验证明该单抗与CEA单抗是两种不同反应区不同性质的单抗。
Different immunohistochemical methods were used to identify the specificity of anti-human colon carcinoma 3 E_4 monoclonal antibody (mouse-derived). A total of 212 specimens were made. It was found that the fluorescence staining of the samples with trypsin digestion was better, and non-specific antigen interference could be ruled out. According to this method, the positive rate of 3E_4 in colon cancer is 100%, positive rate in other adenocarcinomas (pancreas, lung, stomach) is 80%, and the positive rate in normal colon is 0%. In the colon cancer colon, the distribution density was the highest in the periungral membrane and the boundary membrane, the distribution rate was 100%, the upper part of the colon adjacent to the colon was the upper part of the crypt, and there was no cross reaction in the normal colon tissue, indicating that the 3E4 monoclonal antibody is an anti-human colon-associated antibody. . According to a closed test, the monoclonal antibody and CEA monoclonal antibody were proved to be monoclonal antibodies with different properties in two different reaction zones.
