以中华结缕草成熟胚为外植体,研究预处理温度、时间以及不同碳源、氮源、pH值、琼脂浓度、培养温度、激素配比对中华结缕草成熟胚愈伤组织诱导及植株再生的影响。结果表明:成熟胚预处理使用30℃的30%NaOH处理60min可显著提高愈伤组织的诱导率。最佳诱导培养基为MS+酶水解酪蛋白500mg/L、蔗糖3%、葡萄糖2%、琼脂6.5/L,附加2,4-D2mg/L、6-BA0.1mg/L、VB11mg/L、VB21mg/L,灭菌前pH值调至6.0~6.1,最佳培养温度为25℃±2℃。最佳分化培养基为MS,附加6-BA0.3mg/L、NAA0.5mg/L、蔗糖3%、琼脂6.5g/L。
