目的 探索促血小板生成素 (Thrombopoietin ,TPO)分子间相互作用及其存在位置。方法 构建了含有不同TPO区段的酵母双杂交系统融合表达质粒 ,转化酵母菌SFY5 2 6 ,分析 β 半乳糖苷酶活性以推测TPO分子间的相互作用。结果 全长TPO pGBT9与全长TPO pGAD42 4、全长TPO pGAD42 4与TPO(N) pGBT9、TPO(N) pGBT9与TPO(N) pGAD42 4之间有相互作用 ,全长TPO pGBT9与TPO(C) pGAD42 4、TPO(N) pGBT9与TPO(C) pGAD42 4、TPO(C) pGBT9与TPO(C) pGAD42 4之间无相互作用。结论 自然状态下TPO在N端结构区可能存在相互作用 ,C端结构域不参与此过程。
Objective To explore the molecular interaction and location of thrombopoietin (TPO). Methods The yeast two-hybrid system fusion expression plasmid containing different TPO segments was constructed and transformed into yeast SFY5 2 6 to analyze the β-galactosidase activity to speculate the interaction between TPO molecules. Results The full-length TPO pGBT9 interacted with the full-length TPO pGAD42 4, the full-length TPO pGAD42 4 and TPO (N) pGBT9, TPO (N) pGBT9 and TPO ) pGAD42 4, no interaction between TPO (N) pGBT9 and TPO (C) pGAD42 4, TPO (C) pGBT9 and TPO (C) pGAD42 4. Conclusion There may be interaction between TPO and N-terminal domain in nature, and C-terminal domain is not involved in this process.
