目的从微观结构变化考察仙鹿活骨丸对激素性股骨头坏死细胞凋亡影响,为临床应用提供参考。方法给大鼠臀肌注射醋酸强的松龙造成激素型股骨头坏死模型,通过原位细胞凋亡检测、光镜和电镜考察仙鹿活骨丸对激素性股骨头坏死细胞凋亡影响。结果原位细胞凋亡检测分析,发现模型大鼠给仙鹿活骨丸后细胞凋亡数量明显减少;光镜观察可以发现,仙鹿活骨丸可以减少空缺骨陷窝数,降低股骨头组织坏死程度。电镜观察可以发现,仙鹿活骨丸治疗后的骨细胞细胞核染色质和染色体相对均匀,线粒体、高尔基体、内质网等细胞器增多,而糖原溶解与脂肪空泡均较模型对照组减少,因而可提高成骨细胞骨形成的功能,有助于股骨头缺血性坏死修复过程。结论仙鹿活骨丸可以减少激素性股骨头坏死细胞凋亡,改善坏死部位的组织形态学,治疗股骨头坏死。
Objective To investigate the effect of Xianlu Huogu Wan on the apoptosis of steroid-induced necrosis of femoral head from the perspective of microstructural changes, and to provide reference for clinical application. Methods The model of femoral head necrosis was induced by injection of prednisolone acetate into the gluteal muscles of rats. The apoptosis of steroid-induced necrosis of femoral head was investigated by in situ apoptosis assay, light microscope and electron microscopy. RESULTS: The in situ cell apoptosis assay showed that the amount of apoptosis in the model rats was significantly decreased after giving Xianlu Huoqi Pill; light microscope observations showed that Xianlu Huogu Pill can reduce the number of vacated bone lacunae and reduce the femoral head tissue. The degree of necrosis. Electron microscopy showed that after treatment with Xianlu Huogu pill, the nuclear chromatin and chromosomes of bone cells were relatively uniform, and mitochondria, Golgi apparatus, endoplasmic reticulum and other organelles increased, while glycogenolysis and fat vacuolization were decreased compared with the model control group. It can increase the function of osteoblast bone formation and contribute to the repair process of ischemic necrosis of femoral head. Conclusion Xianlu Huogu Pill can reduce the apoptosis of steroid-induced necrosis of femoral head, improve the histomorphology of necrotic area, and treat necrosis of femoral head.
