目的介绍一种快速、有效、高灵敏度的用毛细管气相色谱法测定人血浆中硝苯地平的方法。方法采用SE - 30毛细管色谱柱 (0 .2 4mm× 30m) ,岛津63Ni电子捕获检测器 ,安定作内标。在碱性条件下 ,用乙醚从血浆中提取硝苯地平 ,在气相色谱上直接进样。 结果硝苯地平线性范围为 1~ 10 0 0mg/ml(r =0 .9999) ,最低血浓度检出限 0 .192 pg/ml,绝对回收率 95 %左右。精密度考察硝苯地平日内、日间相对偏差均小于 5 %。硝苯地平在暗室红灯下 ,浓度基本不变。 结论本法适合临床分析研究和应用。
Objective To introduce a rapid, effective and sensitive method for the determination of nifedipine in human plasma by capillary gas chromatography. Methods SE - 30 capillary column (0.24mm × 30m) and Shimadzu 63Ni electron capture detector were used as internal standard. Under basic conditions, nifedipine is extracted from plasma with ether and directly injected on a gas chromatograph. Results The linear range of nifedipine was 1 ~ 100 mg / ml (r = 0.9999). The detection limit of the lowest blood concentration was 0.192 pg / ml and the absolute recovery was about 95%. The precision of nifedipine day, day relative deviation are less than 5%. Nifedipine in darkroom red light, the concentration remained unchanged. Conclusion This method is suitable for clinical analysis and application.
