虽然已知硒是动物及人体的必需元素,但是关于其代谢的知识很不完全。一些研究者应用硫酸铵分级分离、纸上电泳和凝胶过滤等方法分离血液硒结合蛋白,但是由于应用了超生理剂量的硒以及缺乏适宜的蛋白分离方法,以致妨碍了特殊硒蛋白的鉴定。Sandholm于1974年采用了琼脂糖交叉免疫电泳及放射性自显影技术进行血浆硒结合蛋白的分离鉴定,取得较好效果。我们为了研究克山病病区人,畜的血浆硒结合蛋白,按我们实验室条件建立了本法,现介绍如下。
Although selenium is known to be an essential element in animals and humans, knowledge of its metabolism is incomplete. Some researchers use the methods of ammonium sulfate fractionation, electrophoresis on paper and gel filtration to separate blood selenium binding proteins. However, due to the application of ultra-physiological doses of selenium and the lack of suitable protein separation methods, the identification of special selenoproteins is hindered. Sandholm in 1974, the use of agarose cross-immunodetection and autoradiography of plasma selenium binding protein separation and identification, and achieved good results. In order to study the plasma selenium binding protein of human and livestock in Keshan disease area, we established this method according to our laboratory conditions and are introduced as follows.
