目的探讨肿瘤转移抑制基因KiSS1对卵巢癌细胞株HO8910生物学行为的影响。方法含有人KiSS1基因全长cDNA的重组真核表达载体pcDNA3KiSS-1经酶切和测序鉴定正确后,转染卵巢上皮癌细胞株HO8910,观察其KiSS1mRNA表达、细胞生长及增殖能力和细胞侵袭力的变化。结果pcDNA3KiSS1成功转染HO8910细胞,KiSS1mRNA表达阳性,而亲本HO8910细胞为阴性。转染目的基因后细胞生长曲线、软琼脂克隆形成率等与未转染之细胞无显著性差异(P>0.05),但细胞侵袭能力明显下降(P<0.01),穿透Matrigel膜细胞数较未转染组明显减少(P<0.01)。结论KiSS1基因对卵巢上皮癌细胞HO8910的生长、增殖能力无影响,但可抑制其侵袭能力。
Objective To investigate the effect of tumor suppressor gene KiSS1 on the biological behavior of ovarian cancer cell line HO8910. Methods The recombinant eukaryotic expression vector pcDNA3KiSS-1 containing human KiSS1 full-length cDNA was identified by restriction enzyme digestion and sequencing, and then transfected into ovarian epithelial carcinoma cell line HO8910 to observe its KiSS1 mRNA expression, cell growth, proliferation and cell invasiveness Variety. Results The pcDNA3KiSS1 transfected HO8910 cells successfully, KiSS1 mRNA expression was positive, while the parental HO8910 cells were negative. The cell growth curve and the formation rate of soft agar had no significant difference with those of untransfected cells (P> 0.05), but the cell invasion ability decreased significantly (P <0.01). Conclusions The KiSS1 gene has no effect on the growth and proliferation of ovarian epithelial carcinoma HO8910 cells, but inhibits its invasive ability.
