目的研究亚慢性低剂量砷暴露对大鼠的内分泌干扰作用。方法 200只SD大鼠按雄性、雌性均随机分为5组(对照组,0.05、0.1、0.2和0.4μg/m L组),每组雌雄各20只,适应性饲养一周后,经饮水砷染毒分别于110 d和194 d后采用放射免疫法测定血清激素水平。结果不同浓度砷暴露组大鼠血清激素含量发生变化,饲养110 d各组雄鼠雄激素(T)依次为(4.80±4.26)、(4.54±3.31)、(1.27±1.51)、(1.93±2.23)和(4.83±2.31)ng/m L(P=0.05);各剂量组雌鼠甲状腺素(T4)含量依次为:(40.20±7.10)、(40.85±8.66)、(46.49±8.52)、(44.66±11.83)和(26.80±8.12)ng/m L,其中0.4μg/m L组雌鼠T4含量明显降低(P<0.05);各剂量组雌鼠皮质醇(Cort)含量依次为(312.10±10.61)、(333.15±15.73)、(341.13±19.25)、(342.59±25.96)和(314.84±16.76)ng/m L,与对照组相比,染毒组雌鼠Cort均升高,且0.05、0.1和0.2μg/m L组均显著升高(P<0.05)。饲养194 d各种雄鼠T4水平依次为(23.06±5.54)、(33.18±5.29)、(34.23±4.15)、(37.19±4.13)和(39.92±6.27)ng/m L,与对照组比较,各染毒组雄鼠T4均显著升高(P<0.05);各组雌鼠T3水平依次为(0.84±0.12)、(0.89±0.14)、(0.83±0.15)、(0.96±0.16)和(0.75±0.11)ng/m L,其中0.2μg/m L组雌鼠T3显著升高(P<0.05)。其他各染砷组大鼠激素未发生明显变化(P>0.05)。结论亚慢性低剂量砷暴露对SD大鼠具有轻微的内分泌干扰作用。
Objective To investigate the endocrine disrupting effect of subchronic low dose arsenic exposure in rats. Methods 200 male and female SD rats were randomly divided into 5 groups (control group, 0.05, 0.1, 0.2 and 0.4 μg / m L groups), 20 in each group. After one week of adaptability, Serum hormone levels were measured by radioimmunoassay after 110 days and 194 days respectively. The T4 content of female mice in 0.4μg / m L group was significantly decreased (P <0.05) (333.15 ± 15.73), (341.13 ± 19.25), (342.59 ± 25.96) and (314.84 ± 16.76) ng / m L, respectively. Compared with the control group, 0.1 and 0.2μg / m L groups were significantly increased (P <0.05). The levels of T4 in various male rats fed for 194 days were (23.06 ± 5.54), (33.18 ± 5.29), (34.23 ± 4.15), (37.19 ± 4.13) and (39.92 ± 6.27) ng / m L, respectively. Compared with the control group, (P <0.05). The levels of T3 in each group were (0.84 ± 0.12), (0.89 ± 0.14), (0.83 ± 0.15), (0.96 ± 0.16) and 0.75 ± 0.11) ng / m L, in which T3 of 0.2μg / m L group was significantly increased (P <0.05). There was no significant change in other hormones in the other groups (P> 0.05). Conclusion Subchronic low-dose arsenic exposure has a slight endocrine disrupting effect on SD rats.
