用随机扩增的多态性DNA(RAPD)技术对螺原体3个血清组中的4个菌株进行DNA多态性研究。PCR扩增引物选自Operon随机引物试剂盒中I、J、K、L、M、N组的120个引物。4个菌株为236(第Ⅲ血清组),CR1和CH911(第I血清组第2亚组),CCH(第ⅩⅩⅣ血清组)。在120个引物中有21个能在4种菌株DNA模板上扩增出DNA条带,其中在4种模板上皆可扩增出条带的引物有9个。电泳结果显示不同血清组菌株间DNA多态性丰富,而同一血清组中菌株的DNA扩增图谱差异微小。
DNA polymorphisms of 4 strains of 3 serogroups of spirochetes were studied using random amplified polymorphic DNA (RAPD) technique. The PCR amplification primers were selected from 120 primers in I, J, K, L, M, N groups in the Operon Random Primer Kit. Four strains of 23 6 (group Ⅲ serum), CR 1 and CH 91 1 (the first group of serum subgroup 2), CCH (Ⅹ Ⅹ Ⅳ serum group). Of the 120 primers, 21 were capable of amplifying DNA bands on 4 strains of DNA templates, of which 9 were primers capable of amplifying on all 4 types of template. The results of electrophoresis showed that there was abundant DNA polymorphism among strains of different serogroups, but there was no significant difference between the DNA amplifications in the same serogroups.
