目的研究、探讨血小板源性生长因子(plateletderivedgrowthfactor,PDGF)和肝素对人主动脉平滑肌细胞(humanaortasmoothmusclecel,hASMC)增殖、胶原蛋白的合成、分泌以及Ⅰ、Ⅲ型前胶原mRNA表达和转移生长因子-β(transforminggrowthfactor-β,TGF-β)mRNA表达的调节作用。方法3H-TdR,3H-脯氨酸掺入及Northernblot分析。3H-TdR,3H-脯氨酸掺入的比较采用t检验。结果PDGF能促进hASMCDNA的合成和胶原蛋白的合成、分泌;能上调Ⅰ、Ⅲ型前胶原mRNA表达和TGF-βmRNA表达。肝素能抑制hASMCDNA合成和胶原蛋白的合成、分泌,还部分拮抗PDGF促进hASMCDNA合成和胶原蛋白合成、分泌的作用,以及拮抗PDGF上调Ⅰ、Ⅲ型前胶原mRNA表达和TGF-βmRNA表达的作用。结论PDGF能通过上调Ⅰ、Ⅲ型前胶原mRNA表达和TGF-βmRNA表达促进平滑肌细胞(smoothmusclecel,SMC)胶原蛋白的合成、分泌;肝素能抑制SMCDNA和胶原蛋白合成,还能拮抗PDGF的促SMC胶原蛋白合成?
Objective To investigate the effects of platelet derived growth factor (PDGF) and heparin on proliferation, collagen synthesis and secretion of human aorta smooth muscle cells (hASMC) and the expression of type Ⅰ and Ⅲ procollagen mRNA and the expression of transforming growth factor-β (Transforming growth factor-β, TGF-β) mRNA expression regulation. Methods 3H-TdR, 3H-proline incorporation and Northern blot analysis. 3H-TdR, 3H-proline incorporation using t test. Results PDGF could promote the synthesis of hASMCDNA and the synthesis and secretion of collagen, and upregulate the expressions of procollagen type Ⅰ and Ⅲ mRNA and TGF-βmRNA. Heparin can inhibit hASMCDNA synthesis and collagen synthesis, secretion, but also partially antagonize PDGF promote hASMCDNA synthesis and collagen synthesis and secretion, as well as antagonize PDGF upregulated type Ⅰ, Ⅲ procollagen mRNA expression and TGF-βmRNA expression. Conclusion PDGF can promote the synthesis and secretion of collagen in smooth muscle cells (SMCs) by upregulating the expression of procollagen type Ⅰ and Ⅲ mRNA and TGF-β mRNA. Heparin can inhibit the synthesis of SMCDNA and collagen, and also antagonize the promotion of PDGF-stimulated SMC collagen Protein synthesis?
