目的研究低分子肝素(low molecular weight heparin,LMWH)对大鼠大脑皮层神经细胞缺血再灌注损伤的保护作用及其可能机制。方法体外培养新生大鼠大脑皮层神经细胞,建立缺血再灌注模型,MTT法检测细胞活力,Annexin V-FITC、PI双染流式测细胞凋亡率,荧光分光光度计法测定细胞内钙离子浓度。结果低分子肝素可提高缺血再灌注损伤的神经细胞活力,降低细胞凋亡率和细胞内钙离子浓度。结论低分子肝素对缺血再灌注损伤的大鼠大脑皮层神经细胞有保护作用,其机制可能与LMWH降低细胞内钙离子浓度有关。
Objective To investigate the protective effect of low molecular weight heparin (LMWH) on ischemia / reperfusion injury of rat cerebral cortical neurons and its possible mechanism. Methods Cultured neonatal rat cerebral cortical neurons were cultured in vitro to establish a model of ischemia-reperfusion. MTT assay was used to detect the cell viability, Annexin V-FITC and PI double staining flow cytometry. Fluorescence spectrophotometry was used to measure intracellular calcium concentration. Results Low molecular weight heparin increased neuronal viability during ischemia-reperfusion injury and decreased the rate of apoptosis and intracellular calcium concentration. Conclusion Low molecular weight heparin can protect cerebral cortex neurons from ischemia-reperfusion injury in rats. The mechanism may be related to the reduction of intracellular calcium concentration by LMWH.
