目的探讨吡格列酮、多烯磷脂酰胆碱联合治疗对非酒精性脂肪性肝病(NAFLD)大鼠肝细胞内脂质沉积及氧化应激的影响。方法高脂饮食饲养雄性SD大鼠12周,建立大鼠NAFLD模型,采用Seglen胶原酶灌流法分离大鼠肝细胞,常规培养并鉴定。分为正常对照组(NC组)、模型对照组(MC组)、吡格列酮单药组(PG组)、多烯磷脂酰胆碱单药组(PPC组)、联合用药组(PG+PPC组),干预24 h,检测培养液上清中丙氨酸氯基转移酶(ALT)、天冬氨酸转氨酶(AST)水平,肝细胞内丙二醛(MDA)水平、超氧化物歧化酶(SOD)活性、甘油三酯(TG)含量。结果 MC组上清液的ALT、AST水平及肝细胞内MDA、TG含量均较NC组升高(P<0.05),SOD活性较NC组降低(P<0.05);PG组肝细胞内MDA、TG含量较MC组降低(P<0.05),SOD活性较MC组升高(P<0.05);PPC组较MC组上清液中ALT、AST水平及肝细胞内MDA及TG含量均降低(P<0.05),SOD活性较MC组升高(P<0.05);PG+PPC组较MC组上清液ALT、AST水平及肝细胞内MDA、TG含量均降低(P<0.05),SOD活性升高(P<0.05);PPC组较PG组上清液ALT、AST水平及肝细胞内TG含量降低(P<0.05);与PG组、PPC组比较,PG+PPC组肝细胞内TG、MDA水平均降低,SOD活性均升高(P<0.05)。结论吡格列酮和多烯磷脂酰胆碱联用可显著改善NAFLD大鼠肝细胞内脂质沉积及氧化应激水平。
Objective To investigate the effects of combination of pioglitazone and polyene phosphatidylcholine on lipid deposition and oxidative stress in hepatocytes of nonalcoholic fatty liver disease (NAFLD) rats. Methods Male SD rats were fed with high-fat diet for 12 weeks. Rat model of NAFLD was established. The hepatic cells were isolated by Seglen collagenase perfusion and cultured and identified routinely. They were divided into normal control group (MC group), model control group (MC group), pioglitazone single group (PG group), polyene phosphatidylcholine single group (PPC group) (ALT), aspartate aminotransferase (AST), malondialdehyde (MDA) in hepatocytes, superoxide dismutase (SOD) ) Activity, triglyceride (TG) content. Results The levels of ALT and AST and the levels of MDA and TG in hepatocytes in MC group were higher than those in NC group (P <0.05), while the activities of SOD in MC group were lower than those in NC group (P <0.05) (P <0.05), while the activity of SOD in the PPC group was lower than that in the MC group (P <0.05). The levels of ALT and AST and the content of MDA and TG in the PPC group were lower than those in the MC group (P <0.05). The activity of SOD in the PG + PPC group was lower than that in the MC group (P <0.05), while the levels of ALT and AST, (P <0.05). Compared with PG group and PPC group, the level of ALT, AST and the level of TG in hepatocytes in PPC group were lower than that in PG group (P <0.05) The levels were decreased, SOD activity were increased (P <0.05). Conclusion Combination of pioglitazone and polyene phosphatidylcholine can significantly improve lipid deposition and oxidative stress in NAFLD rats.
