目的:探讨流式细胞术三色荧光/侧向角散射设门(CD45/SSC-Gating)与散射光设门(FSC/SSC-Gating)两种设门方法对乳腺癌前哨淋巴结淋巴细胞亚群分析的差异及其临床应用价值。方法:将前哨淋巴结制备成单细胞混悬液,用CD45/SSC、FSC/SSC两种方法设门,流式细胞仪直接计数前哨淋巴结中淋巴细胞亚群的百分比,并用统计学方法分析比较结果。结果:FSC/SSC设门组中总淋巴细胞的百分比(TL)为(79·42±5·02)%,CD45/SSC设门组中TL为(97·66±4·60)%(P<0·05)。同时两个设门组的CD3+T,CD4+T,CD8+T测定值比较,差异亦有统计学意义(P<0·05)。结论:在分析乳腺癌前哨淋巴结淋巴细胞亚群时,CD45/SSC设门优于FSC/SSC设门。
OBJECTIVE: To investigate the effects of flow cytometry (CD45 / SSC-Gating) and FSC / SSC-Gating methods on breast cancer SLNs Analysis of the difference and its clinical value. Methods: Sentinel lymph nodes were prepared as single-cell suspensions. CD45 / SSC and FSC / SSC methods were used to determine the percentage of lymphocyte subsets in sentinel lymph nodes by flow cytometry. Statistical analysis was performed to compare the results . Results: The percentage of total lymphocytes (FS) in FSC / SSC group was (79.42 ± 5.22)% and that in CD45 / SSC group was (97.66 ± 4.60)% (P <0 · 05). At the same time, the difference of CD3 + T, CD4 + T and CD8 + T between the two gating groups was also statistically significant (P <0.05). Conclusion: CD45 / SSC gate is superior to FSC / SSC gate in analyzing sentinel lymph node lymphocyte subsets in breast cancer.
