目的:观察脂联素(adiponectin,APN)水平对心肌脂联素受体1(adiponectin receptors 1,AdipoR1)表达的影响;探讨胰岛素抵抗(insulin resistance,IR)是否是APN影响心肌AdipoR1表达的机制之一。方法:观察对象分为5组,其中野生型C57BL小鼠分为:对照组(CON组)和IR组,APN~(-/-)小鼠分为:APN~(-/-)组(KO组),APN~(-/-)+IR组(KIR组)和APN~(-/-)+IR+APN干预组(APN组)。IR组、KIR组、APN组小鼠给予高脂饲料诱导IR模型,其中APN组小鼠还给予重组APN腹腔注射。干预12周后取血,用血糖仪检测小鼠空腹血糖(fasting plasma glucose,FPG)、ELISA法检测血清空腹胰岛素(fasting insulin,FINS)及APN水平,并计算IR指数(HOMA-IR)。取小鼠心肌组织,采用免疫组织化学法、Western blot法、PCR法检测心肌AdipoR1的表达水平。结果:与CON组比较,KO组和IR组血清APN水平降低、心肌AdipoR1蛋白表达降低,IR组HOMA-IR水平升高。与KIR组比较,APN组血清APN水平升高、心肌AdipoR1蛋白升高、HOMA-IR水平降低。小鼠心肌AdipoR1与血清APN呈正相关,与FINS、HOMA-IR呈负相关。结论:APN可上调心肌AdipoR1的表达。IR可导致心肌AdipoR1表达下调。APN可通过改善IR起到上调心肌AdipoR1表达的作用。
OBJECTIVE: To investigate the effect of adiponectin (APN) on the expression of adiponectin receptors 1 (AdipoR1), and to explore whether insulin resistance (IR) is the mechanism by which APN affects the expression of AdipoR1 in myocardium one. Methods: The subjects were divided into five groups: wild-type C57BL mice were divided into control group (CON group) and IR group. APN ~ (- / -) mice were divided into APN group APN ~ (- / -) + IR group (KIR group) and APN ~ (- / -) + IR + APN intervention group (APN group). IR group, KIR group, APN group were given high fat diet induced IR model, in which APN mice were also given recombinant APN intraperitoneal injection. After 12 weeks of intervention, blood samples were taken for fasting plasma glucose (FPG), fasting insulin (FINS) and APN levels were detected by ELISA, and HOMA-IR was calculated. Myocardial tissue was harvested and the expression of AdipoR1 in myocardium was detected by immunohistochemistry, Western blot and PCR. Results: Compared with CON group, serum APN level decreased, myocardial AdipoR1 protein level decreased and HOMA-IR level increased in IR and KO groups. Compared with KIR group, APN increased serum APN levels, myocardial AdipoR1 protein increased, HOMA-IR levels decreased. Myocardial AdipoR1 was positively correlated with serum APN and negatively correlated with FINS and HOMA-IR. Conclusion: APN can up-regulate the expression of AdipoR1 in myocardium. IR can lead to myocardial AdipoR1 expression down regulation. APN plays a role in up-regulation of myocardial AdipoR1 expression by improving IR.
