以甘蔗野生近缘植物斑茅(S.arundinaceum Retz)为外源DNA供体材料,栽培桂糖11号为受体材料,用“酚/SDS改良法”提取供体DNA.利用基因枪转导法把浓度为1μg/μl的供体DNA转导入受体细胞并整合至受体染色体上获得表达。从再生植株中筛选获得3个与受体在农艺性状表现有差异的转异再生杂种株系,并通过叶片过氧化酶同工酶分析得以初步认定.这种人工种质新材料可供进一步育种研究。
Using S.arundinaceum Retz as exogenous DNA donor material and Guiguang 11 as acceptor material, the donor DNA was extracted by phenol / SDS modified method. Transfection of donor DNA at a concentration of 1 μg / μl using gene gun transduction into recipient cells and integration into recipient chromosomes resulted in expression. Three regenerated hybrid lines with different agronomic traits were screened from regenerated plants and were initially identified by leaf peroxidase isoenzyme analysis. This new artificial germplasm material is available for further breeding research.
