目的:探讨结直肠癌肝转移(CRLM)组织中TGF-β1、Smad7表达和树突状细胞(DC)的变化及临床意义。方法:采用免疫组化法检测TGF-β1、Smad7、CD1α(未成熟DC标志)和CD83(成熟DC标志)在CRLM组织和CRLM术后切缘阴性的正常肝组织中的表达,分析CRLM组织中以上指标表达之间的关系以及与原发灶特征的关系。结果:CRLM组织中TGF-β1、Smad7、和CD1α的表达明显高于正常肝组织组(均P<0.05),且CRLM组织中彼此表达之间呈正相关关系(r=0.455、r=0.623、r=0.448,均P<0.05);CD83在CRLM组织中基本为全阴性表达,而在正常肝组织组中仅2例呈弱阳性表达。TGF-β1、Smad7、和CD1α的表达均与CRLM患者原发灶的肿瘤浸润深度、有无淋巴结转移有关(均P<0.05),而与原发灶肿瘤分化程度无关(P>0.05)。结论:TGF-β1、Smad7α的高表达及未成熟DC数量的增加可能共同促进CRLM的发生、发展。
Objective: To investigate the changes of TGF-β1, Smad7 and dendritic cells (DCs) in colorectal cancer liver metastasis (CRLM) and its clinical significance. Methods: Immunohistochemistry was used to detect the expression of TGF-β1, Smad7, CD1α (immature DC marker) and CD83 (mature DC marker) in CRLM tissues and normal liver tissues with negative margins after CRLM. The above indicators of the relationship between the expression and the relationship between the characteristics of the primary tumor. Results: The expressions of TGF-β1, Smad7 and CD1α in CRLM tissues were significantly higher than those in normal liver tissues (all P <0.05), and there was a positive correlation between CRLM expression and CRLM expression (r = 0.455, r = 0.623, r = 0.448, all P <0.05). CD83 expression was almost completely negative in CRLM tissues and only weakly positive in 2 of the normal liver tissues. The expression of TGF-β1, Smad7 and CD1α were correlated with the depth of tumor invasion and lymph node metastasis (P <0.05) in primary tumor of CRLM patients, but not with the degree of primary tumor differentiation (P> 0.05). Conclusion: The high expression of TGF-β1 and Smad7α and the increase of the number of immature DC may promote the occurrence and development of CRLM.
