目的建立反相高效液相色谱法测定枸杞中芦丁含量的方法。方法色谱分析条件Shim-packVP-ODS色谱柱(250mm×4.6mm,5μm),以乙腈∶水∶冰乙酸(5∶94.5∶0.5)为流动相A,乙腈∶水∶冰乙酸(70∶29.5∶0.5)为流动相B,采用梯度洗脱,流速0.5ml/min,检测波长342nm,柱温30℃。结果芦丁线性范围为10~100μg/ml(相关系数为r=0.9998),平均回收率(n=5)为99.0%;不同产地枸杞样品中芦丁含量存在差异,随保存时间的延长,芦丁含量明显下降。结论本方法重现性好,适用于测定枸杞中芦丁的含量。
Objective To establish a method for the determination of rutin in wolfberry by RP-HPLC. Methods The chromatographic conditions were on a Shim-pack VP-ODS column (250 mm×4.6 mm, 5 μm) with acetonitrile:water:glacial acetic acid (5:94.5:0.5) as the mobile phase A, acetonitrile:water:glacial acetic acid (70:29.5: 0.5) for mobile phase B, gradient elution, flow rate 0.5 ml/min, detection wavelength 342 nm, column temperature 30°C. Results The linear range of rutin was 10~100μg/ml (the correlation coefficient was r=0.9998), and the average recovery rate (n=5) was 99.0%. Rutin content was different in the samples from different regions, and with the extension of preservation time, Ding content decreased significantly. Conclusion The method has good reproducibility and is suitable for the determination of rutin in loquat.
