目的:通过实验研究绞股蓝总皂苷对2型糖尿病大鼠游离脂肪酸代谢的影响,探讨绞股蓝总皂苷改善2型糖尿病大鼠胰岛素抵抗相关机制。方法:将实验用SD大鼠建造为2型糖尿病模型,分别给予药物灌服及普通饲料喂养4周。采用荧光定量PCR法检测各组大鼠肝脏及脂肪组织中PPAR-a mRNA;血清学测定血清中游离脂肪酸(NEFA)、空腹血糖(FPG)、甘油三脂(TG)、胰岛素(INS)的含量,根据血糖值及胰岛素的测定结果计算出胰岛素抵抗指数;采用胰腺β细胞HE染色及细胞凋亡检测胰岛细胞凋亡。结果:绞股蓝总苷高、中、低剂量能提高PPAR-a mRNA的表达;与模型组比较有所改善,绞股蓝总苷高剂量能明显降低糖尿病大鼠的血糖、血脂、游离脂肪酸;绞股蓝总苷高、中、低剂量动物胰腺B细胞可见凋亡,绞股蓝总苷高、中、低剂量均能增强糖尿病大鼠的胰岛素水平,增强对胰岛素的敏感性,减小抵抗指数。结论:绞股蓝总皂苷可以通过提高PPAR-a mRNA的表达,明显降低糖尿病大鼠游离脂肪酸浓度,从而增强胰岛素敏感性、减少胰岛素抵抗。
Objective: To study the effect of gypenosides on the free fatty acid metabolism in type 2 diabetic rats and to explore the mechanism of gypenosides improving insulin resistance in type 2 diabetic rats. Methods: Experimental SD rats were used as model for type 2 diabetes mellitus. The rats were fed with drugs and normal diet for 4 weeks respectively. The levels of PPAR-a mRNA in liver and adipose tissue were detected by real-time PCR. Serum levels of free fatty acids (NEFA), fasting blood glucose (FPG), triglyceride (TG) and insulin The insulin resistance index was calculated according to the blood glucose level and insulin measurement results. The islet cell apoptosis was detected by HE staining and apoptosis of pancreatic β cells. Results: High, medium and low doses of gypenosides could increase the expression of PPAR-a mRNA. Compared with the model group, Gynostemma pentaphyllum total glycosides high dose could significantly reduce blood glucose, blood lipids and free fatty acids in diabetic rats; High, medium and low doses of pancreatic B cells showed apoptosis, gypenosides glycosides high, medium and low doses can enhance the insulin level in diabetic rats, enhance insulin sensitivity, reduce the resistance index. Conclusion: Gypenosides can increase the expression of PPAR-a mRNA, significantly reduce the concentration of free fatty acids in diabetic rats, so as to enhance insulin sensitivity and reduce insulin resistance.
