目的探讨人卵巢癌组织差异表达基因,揭示卵巢癌发生、发展的分子机制。方法人卵巢癌组织标本2例,均经病理确诊为低分化浆液性乳头状囊腺癌,临床诊断为卵巢癌Ⅲ期,选用2例正常卵巢组织为对照。提取卵巢组织中mRNA,提取并纯化卵巢癌组织中总RNA,应用cDNA基因芯片技术对总共588个基因进行差异表达基因检测。结果共筛选出差异表达基因44条,其中11条表达上调(包括癌基因c-erbB2、neu、c-fos、c-mycproto-oncogenes、HER2receptor等)、33条表达下调(包括RAR、MMP18、MMP19、p21、DNA-PK等)。结论揭示了人卵巢癌组织中差异表达基因,有望为早期诊断卵巢癌及判断预后提供理想标志物。
Objective To investigate differentially expressed genes in human ovarian cancer and to reveal the molecular mechanism of the occurrence and development of ovarian cancer. Methods Two cases of human ovarian cancer tissues were diagnosed as poorly differentiated serous papillary cystadenocarcinoma by pathology. The clinical diagnosis was stage Ⅲ of ovarian cancer. Two cases of normal ovarian tissue were selected as control. The mRNA of ovarian tissue was extracted, and the total RNA was extracted and purified from ovarian cancer tissues. A total of 588 genes were detected by cDNA microarray. Results A total of 44 differentially expressed genes were screened, of which 11 were upregulated (including oncogenes c-erbB2, neu, c-fos, c-myc proto-oncogenes and HER2receptor) and 33 were downregulated (including RAR, MMP18 and MMP19 , P21, DNA-PK, etc.). Conclusions revealed that differentially expressed genes in human ovarian cancer tissue are expected to provide an ideal marker for early diagnosis of ovarian cancer and prognosis.
