目的建立蓖麻毒素和相思子毒素蛋白质芯片检测方法。方法制备并纯化蓖麻毒素和相思子毒素单克隆抗体。采用竞争免疫法检测抗原,建立检测蓖麻毒素和相思子毒素的蛋白质芯片检测方法。应用蛋白质芯片检测两种毒素的模拟样品。结果应用蛋白质芯片检测蓖麻毒素、相思子毒素和两种毒素混合的模拟样品时,在其相应区域均没有荧光信号,结果为阳性,与实验设计相符。结论已成功建立了蓖麻毒素和相思子毒素蛋白质芯片检测方法,该方法具有广阔的应用前景。
Objective To establish a method for the detection of ricin and acacia protein chip. Methods Monoclonal antibodies against ricin and acacci were prepared and purified. Antigen was detected by competitive immunoassay and protein chip detection method was established to detect ricin and acacia. Simulate samples of both toxins using a protein chip. Results When using the protein chip to detect ricin, acacia and mixed samples of two kinds of toxins, there was no fluorescence signal in the corresponding region, and the result was positive, which accorded with the experimental design. Conclusion The detection method of ricin and acacia protein chip has been successfully established. The method has broad application prospects.
