目的:建立枸杞叶中芦丁的含量测定方法。方法:采用Diamonsil C18(250mm×4.6mm,5μm)色谱柱,流动相为甲醇-0.5%磷酸溶液,梯度洗脱,流速为1.0mL·min-1,检测波长为358nm。结果:芦丁进样浓度在50.7~404.8μg·mL-1范围内线性关系良好(r=0.9999);低、中、高浓度的平均回收率(n=3)分别为101.8%(RSD=1.8%),98.6%(RSD=1.5%),97.9%(RSD=0.7%)。结论:本法准确、可靠,灵敏度高,可作为枸杞叶中芦丁的定量分析方法。
Objective: To establish a method for the determination of rutin in loquat leaves. Methods: A Diamonsil C18 (250mm×4.6mm, 5μm) column was used. The mobile phase consisted of methanol-0.5% phosphoric acid solution with gradient elution. The flow rate was 1.0mL·min-1. The detection wavelength was 358nm. RESULTS: The linearity of the rutin injection concentration ranged from 50.7 to 404.8 μg·mL-1 (r=0.9999). The average recoveries of the low, medium and high concentrations (n=3) were 101.8% (RSD=1.8). %), 98.6% (RSD = 1.5%), 97.9% (RSD = 0.7%). Conclusion: This method is accurate, reliable and has high sensitivity. It can be used as a quantitative analysis method for rutin in loquat leaves.
