目的探讨神经酰胺(Cer)对小鼠皮层神经元噻唑蓝(MTT)代谢率的影响。②方法以原代培养的26只小鼠大脑皮层神经元为模型,观察Cer不同浓度(0,50,100,200,500,1000,2000,5000μmol/L),不同的作用时间(4,8,12,16,24,36h)对其MTT代谢率的影响。③结果MTT代谢率随着Cer浓度的增加及作用时间的延长明显下降(t=2.08~2.14,P<0.05)。④结论Cer可损伤神经元线粒体功能,从而导致神经元死亡。
Objective To investigate the effect of ceramide (Cer) on the metabolic rate of thiazolyl blue (MTT) in mouse cortical neurons. Methods The cerebral cortical neurons of 26 mice were cultured in different concentrations (0, 50, 100, 200, 500, 1000, 2000 and 5000μmol / L) , 12,16,24,36 h) on its MTT metabolic rate. Results The metabolic rate of MTT decreased significantly with the increase of Cer concentration and prolongation of action time (t = 2.08-2.14, P <0.05). Conclusion Cer can damage the neuronal mitochondrial function, leading to neuronal death.
