This article describes the frozen storage of semen from tilapia in Mozambique when it is cryogenically protected with 5% methanol and 15% milk powder. This method of sperm freezing thaw sperm motility is about 5% or 10% of dimethyl sulfoxide (DMSO) instead of methanol as a cryogenic protective agent twice. This article also describes in detail the optimum concentration of cryoprotectant, the optimum temperature for cooling and thawing, and the effect of dilution and equilibration time on sperm. A fertilization test was also reported: 64.3% ± 34.2% of frozen-stored spermatozoa fertilized embryos developed normally to the embryo confinement compared to 57.5 ± 10.5% in the control group without freezing.
