为了探讨生后大鼠脑室下层神经元前体细胞离体培养时的神经活性物质特征和形态学特征 ,本研究以细胞特异性的抗体进行免疫细胞化学反应并测量了这些细胞的形态学指标胞体平均直径和胞体椭圆率 (胞体最小径与最大径的比值 )。取脑室下层细胞接种于覆有多聚赖氨酸的 2 4孔培养板 ,培养液为 Neurobasal Medium( Gibco) /B2 7,培养条件为 5 % CO2 ,3 7℃。结果如下 :在培养 1d时 ,绝大多数 (大于 90 % )脑室下层源细胞呈 β-3型微管蛋白阳性 ,且这些细胞也都表达多唾液酸神经粘连分子。此时 ,大多数β-3型微管蛋白阳性细胞胞体呈椭圆形 (胞体平均直径为 8.42± 1.0 3μm;胞体椭圆率为 0 .5 7± 0 .12 ) ,且在其两极各长出一短的、无分支的突起 ;并有近 2 0 %者为球形无突起细胞 (胞体平均直径为 7.2 0± 1.0 4μm) ,且它们大多呈细胞与细胞相连接状态。随着培养时间的延长 ,这些细胞亦呈上述椭圆形有突起样细胞变化 ,但仍可见它们呈串珠样连接。培养至第 3~5 d,这些脑室下层源细胞的体积增大 (胞体平均直径为 9.0 7± 1.0 7μm)、突起较前增长但仍几无分支。免疫细胞化学反应显示 ,此时培养中的脑室下层源细胞仍大多呈β-3型微管蛋白阳性和多唾液酸神经粘连分子阳性。至培养的第 7d,这些β-3型微管蛋白阳性
