探讨卵巢癌细胞凋亡过程相关基因及其蛋白表达的变化 ,为解决卵巢癌化疗耐药提供理论依据。方法 :应用不同浓度顺铂处理HO - 8910细胞 ,HE染色观察细胞形态改变 ,琼脂糖凝胶电泳观察DNA降解 ,SP免疫组化染色观察细胞凋亡过程中P53、Bcl - 2蛋白表达的变化。结果 :由于使用顺铂 ,HO - 8910细胞呈凋亡改变 ,DNA琼脂糖凝胶电泳呈典型的凋亡特征。在细胞凋亡的同时 ,P53蛋白表达逐渐升高 ,Bcl- 2蛋白表达逐渐降低 ,二者的变化分别和凋亡细胞数呈正、负相关性。结论 :顺铂通过细胞内P53、Bcl - 2蛋白表达的变化 ,诱导人卵巢癌细胞凋亡是其抗肿瘤的机制之一。卵巢癌细胞对顺铂耐药的产生可能和P53、Bcl- 2蛋白表达有关。
To investigate the changes of apoptosis related genes and their protein expression in ovarian cancer cells and to provide a theoretical basis for solving the chemotherapy resistance in ovarian cancer. Methods: HO - 8910 cells were treated with different concentrations of cisplatin. The morphological changes of cells were observed by HE staining. DNA degradation was observed by agarose gel electrophoresis. The expression of P53 and Bcl - 2 protein was observed by SP immunohistochemical staining. Results: HO - 8910 cells showed apoptotic changes due to the use of cisplatin, and DNA agarose gel electrophoresis showed typical apoptotic features. At the same time of apoptosis, the expression of P53 protein gradually increased, while the expression of Bcl-2 protein gradually decreased. The changes of them both correlated positively with the number of apoptotic cells. Conclusion: Cisplatin can induce apoptosis of human ovarian cancer cells through the changes of P53 and Bcl - 2 protein expression in cells. Ovarian cancer cells resistant to cisplatin may be related to the expression of P53 and Bcl-2 protein.
