目的建立一种定量检测去白细胞悬浮红细胞中残留白细胞的新方法。方法使用ADAM-r白细胞计数系统对100份去白细胞悬浮红细胞中白细胞残留量进行检测,同时用流式细胞仪对上述100份标本进行平行检测;并选用3个浓度白细胞残留量的去白细胞悬浮红细胞对ADAM-r白细胞计数系统白细胞残留量检测进行重复性试验。结果 ADAM-r白细胞计数系统和流式细胞仪2种方法检测去白细胞悬浮红细胞中残留白细胞数的相关系数r=0.921,P<0.001,2种方法检测结果具有统计学关联;重复性试验中3个浓度白细胞残留量的去白细胞悬浮红细胞白细胞残留量检测CV值分别为19.1、4.1、3.8。结论可以使用ADAM-r白细胞计数系统定量检测去白细胞悬浮红细胞中的残留白细胞。
Objective To establish a new method for quantitative determination of leukocytes in leukocyte-depleted erythrocytes. Methods The leukocyte count in 100 leukocyte-depleted erythrocytes was detected by ADAM-r leukocyte counting system. The above 100 samples were detected by flow cytometry in parallel. Three leukocyte-depleted leukocyte-depleted erythrocytes The ADAM-r leukocyte count system leukocyte residue was tested for repeatability. Results The correlation coefficients of ADAM-r leucocyte counting system and flow cytometry in determining the number of leucocytes in leukocyte-depleted erythrocytes were 0.921, P <0.001.
