目的:探讨经载体介导的RNA干涉对人核受体NR5A2表达抑制的可行性。方法:设计并构建人nr5a2特异siRNA表达载体pSilencer1.0-U6-nr5a2,经脂质体介导转入肝癌细胞BEL-7402,RT-PCR分析nr5a2表达情况。结果:瞬时转染重组载体pSilencer1.0-U6-nr5a2的BEL-7402细胞其nr5a2的表达明显受抑,抑制效率可达70%。结论:siRNA表达载体介导的RNA干涉可在细胞水平有效抑制EEL-7402细胞nr5a2的表达。
OBJECTIVE: To investigate the feasibility of inhibiting the expression of NR5A2 by nuclear vector mediated by RNA interference. Methods: The human nr5a2 specific siRNA expression vector pSilencer1.0-U6-nr5a2 was designed and constructed and transfected into hepatoma BEL-7402 by liposome. The expression of nr5a2 was analyzed by RT-PCR. Results: The expression of nr5a2 in BEL-7402 cells transiently transfected with pSilencer1.0-U6-nr5a2 was significantly inhibited and the inhibitory efficiency was up to 70%. Conclusion: RNA interference mediated by siRNA expression vector can effectively inhibit the expression of nr5a2 in EEL-7402 cells at the cellular level.
